چکیده

The Agilent 1260 Infi nity Analytical Supercritical Fluid Chromatography (SFC) System provides LC-like sensitivity and UHPLC power range. It is a pressurized system that has a fi xed-loop autosampler. The injection loops come in different sizes to perform fi xed-loop injections. Injecting a volume smaller than the loop volume is possible using the partial loop-fi ll injection method where the loop is fi lled only partially with sample. This Technical Overview shows the usability of the partial loop-fi ll method to inject variable volumes. The method provides area and retention time RSD (%) of less than 3.5% and 0.1% respectively and results in a linear response for increasing injector volumes. Agilent Application Solution Transfer of a USP method for tolazamide from normal phase HPLC to SFC using the Agilent 1260 Infi nity Hybrid SFC/UHPLC System

Normal phase liquid chromatography methods often have long run times and involve environmentally toxic/costly solvents. Supercritical fl uid chromatography (SFC) methods on the other hand, are faster, inexpensive, and eco-friendly. SFC involves the use of low viscosity supercritical carbon dioxide, which can be operated at fl ow rates up to three times higher than liquid chromatography (LC) wit...
